Molecular Detection of Novel Genetic Variants Associated to Anaplasma ovis among Dromedary Camels in Iran

چکیده مقاله:
To the best of our knowledge, little information is available regarding the presence of Anaplasma species in camels in Iran. This study sought to investigate the presence of Anaplasma species by microscopy and polymerase chain reaction (PCR) assays in 100 healthy dromedaries (Camelus dromedarius) arriving for slaughter. The microscopic examination of Giemsa-stained blood films revealed that Anaplasma like structures could be identified in the erythrocytes of two blood smears. To confirm the presence of and to identify the species of Anaplasma spp., a PCR technique was performed using primers amplifying a 750 bp fragment of the 16S rRNA gene of Anaplasma and the PCR products were analyzed by sequencing. The nucleotide sequence was compared to the sequences available in GenBank using the Basic Local Alignment Search Tool (BLAST). According to the results, the sequences of two 16S rRNA PCR products clearly fit within the Anaplasma genus in the family Anaplas mataceae. In this study, phylogenetic analysis using the 16S rRNA gene sequences revealed that two sequences obtained from monophyletic clusters included Anaplasma ovis (A. ovis). The obtained sequences had 99.6-100% similarity with previously published 16S rRNA gene sequences. This study aimed to evaluate the presence of novel genetic variants associated to A. ovis in dromedaries in the world. Further studies are recommended to establish the vector(s), as well as the veterinary and medical significance of these apparently novel variants in Iran.To the best of our knowledge, little information is available regarding the presence of Anaplasma species in camels in Iran. Molecular Detection of Anaplasma marginale and Anaplasma ovis (Rickettsiales: Anaplasmataceae) in Ixodid Tick Species in Iran
The present study was conducted as the first molecular detection of Anaplasma species in tick samples based on the sequencing of major surface proteins 4 (msp4) gene fragments in different parts of Iran. A total of 130 tick specimens were collected from Hormozgan, Lorestan, and Guilan, Iran, within 2015 to 2017. Hyalomma asiaticum, Hyalomma dromedarii</em...
متن کاملMolecular Detection of Bovine Leukocytic Anaplasma Species in Isfahan, Iran
Background: A. bovis and A. phagocytophilium are leukocytotropic agents of bovine anaplasmosis. They are obligate intracellular bacteria that can infect and cause Anaplasmosis in human and animals. Therefore, this study was carried out to detect A. bovis and A. phagocytophilum in naturally infected dairy cattle in Isfahan using molecular techniques. Materials and Methods: In this study a tot...

This study was carried out to determine the presence and frequency of Anaplasma ovis and Anaplasma marginale in sheep and dairy cattle in West-Azerbaijan province, Iran. A total number of 200 blood samples were randomly collected via the jugular vein from apparently healthy cattle (100) and sheep (100). The extracted DNA from blood cells was screened using genus-specific (Anaplasma...
متن کاملMolecular detection of novel Anaplasmataceae closely related to Anaplasma platys and Ehrlichia canis in the dromedary camel (Camelus dromedarius).
Serological surveys have confirmed Anaplasma marginale and Anaplasma phagocytophilum infections in dromedary camels, but molecular surveys and genetic characterisation of camel-associated Anaplasma species are lacking. In this study, we detected tick-borne Anaplasmataceae in 30 of 100 (30%) healthy dromedary camels screened using a combined 16S rRNA-groEL PCR-sequencing approach. Nucleotide seq...

INTRODUCTION Little information is currently available regarding the presence of Anaplasma species in North African dromedaries. To fill this gap in knowledge, the prevalence, risk factors, and genetic diversity of Anaplasma species were investigated in Tunisian dromedary camels. METHODOLOGY A total of 226 camels from three different bioclimatic areas were sampled and tested for the presence ...
